dc.description.abstract | [ES] La presente Tesis Doctoral supone un paso más en esta línea de investigación dedicada a la búsqueda de una terapia celular efectiva para la paliación de procesos neurodegenerativos. Como objetivos concretos se proponen: - Realizar trasplantes de médula ósea sana en ratones mutantes y silvestres utilizando medios de ablación de la médula de los animales receptores tanto físicos como químicos. Comparar ambos métodos de ablación. - Realizar un seguimiento de la incorporación progresiva de las células trasplantadas en los animales receptores para determinar el éxito del trasplante. - Analizar los mecanismos de plasticidad responsables de la integración de las células derivadas de la médula ósea en el encéfalo. - Estudiar la influencia de la mutación PCD (Purkinje Cell Degeneration) en la incorporación de elementos de la médula ósea. Realizar un estudio cuantitativo de esta integración en el cerebelo y en el bulbo olfativo. - Estudiar el comportamiento y la fisiología de los animales trasplantados, con especial hincapié en ciertas funciones motoras y olfativas. - Estudiar posibles efectos secundarios de la metodología del trasplante de médula ósea en las regiones neurogénicas rostrales del encéfalo del ratón. - Realizar un estudio aproximativo de los niveles de estrés oxidativo y de neuroprotección en la sangre, por medio del análisis de los valores de glutation e IGF-I en los grupos experimentales estudiados. Como resultado de todos estos objetivos parciales, y como objetivo general de esta Tesis Doctoral, se propone conseguir una recuperación estructural y funcional en dos regiones encefálicas afectadas de neurodegeneración: el bulbo olfativo y el cerebelo. La consecución de estos objetivos supondrá un mayor conocimiento de la aplicación de las células madre de la médula ósea como herramienta para la paliación de enfermedades neurodegenerativas y el desarrollo de futuras terapias regenerativas. | es_ES |
dc.description.abstract | [EN] This thesis is a step in this line of research dedicated to finding effective cellular therapy for palliation of neurodegenerative processes. As specific objectives are proposed: - Make healthy bone marrow transplants in mice using mutant and wild media marrow ablation of recipient animals both physical and chemical. Compare the two methods of ablation. - To monitor the progressive incorporation of transplanted cells in recipient animals to determine the success of transplantation. - Analyze the plasticity mechanisms responsible for the integration of cells derived from bone marrow in the brain. - To study the influence of the mutation PCD (Purkinje Cell Degeneration) in the incorporation of elements of the bone marrow. Conduct a quantitative study of this integration in the cerebellum and olfactory bulb. - To study the behavior and physiology of the transplanted animals, with special emphasis on certain features and olfactory motor. - Explore possible side effects of the methodology of bone marrow transplantation in the rostral neurogenic regions of the brain of the mouse. - Conduct a study approximate the levels of oxidative stress and neuroprotection in the blood, through the analysis of the values of glutathione and IGF-I in the experimental groups studied. As a result of all these sub-goals and overall objective of this thesis, aims to achieve a structural and functional recovery in two brain regions affected by neurodegeneration: the olfactory bulb and cerebellum. Achieving these objectives will mean a greater knowledge of the application of stem cells from bone marrow as a tool for the alleviation of neurodegenerative diseases and the development of future regenerative therapies. | |
