Caracterización de las proteínas Mtl2p y Wsc1p, como posibles sensores en la pared celular de Schizosaccharomyces pombe

Resumen
[ES] Las células de Schizosaccharmomyces pombe presentan una forma cilíndrica y crecen por los polos hasta alcanzar un tamaño determinado, entonces se dividen mediante la formación de un septo transversal en la región media de la célula, que separa a las dos células hijas. En cada uno de estos estados de polarización, la síntesis de nueva pared celular es necesaria para mantener tanto la forma como la integridad de la célula. En nuestro grupo estudiamos la regulación de la síntesis del (1,3)-glucano, que es el componente mayoritario de la pared. La enzima encargada de la biosíntesis de este polímero es la (1,3)-glucán sintasa (GS) que es una enzima multimérica formada por un componente catalítico y un componente regulador (la GTPasa Rho1p). El objetivo de este trabajo consiste en la caracterización de nuevos componentes que actúen por encima de la ruta Rho1p/Pck1p/Pck2p que regula la integridad celular en Schizosaccharomyces pombe.
Las proteínas Mtl2p y Wsc1p de S. pombe presentan características estructurales similares a los "sensores" de S. cerevisiae Wsc1p y Mid2p. Un único dominio transmembrana, que conecta las proteínas con la larga región extracelular, un dominio citoplasmático en el extremo carboxilo terminal, y un ectodominio periplásmico rico en serinas y treoninas. Se localizan en la superficie celular pero de manera diferente, Mtl2p se distribuye por toda la periferia de la célula y en el septo. Wsc1p se localiza de forma polarizada y aparece más concentrada en "pequeños gránulos" en los polos de la célula y participan en la regulación de la integridad celular donde desarrollan una función esencial. La deleción simultánea de mtl2+ y wsc1+ es letal. Mtl2p y Wsc1p participan en la activación de Rho1p y los dos interaccionan genéticamente con Rgf1p (un GEF de Rho1p). Wsc1p está involucrada en la regulación de la biosíntesis de b(1,3)-glucano y probablemente también en la de a(1,3)-glucano. Las células que sobreexpresan Wsc1p presentan un incremento general en la cantidad total de pared celular y deposiciones aberrantes de Calcoflúor. Además, Wsc1p es necesario para la activación de Rgf1p al menos en condiciones de estrés. [EN] Schizosaccharmomyces pombe cells have a cylindrical shape and grow through the poles to reach a certain size, then divide by forming a transverse septum in the middle region of the cell, which separates the two daughter cells. In each of these states of polarization, the synthesis of new cell wall is necessary to maintain both the shape and integrity of the cell. In our group we study the regulation of the synthesis of (1,3)-glucan, which is the main component of the wall. The enzyme responsible for the biosynthesis of this polymer is (1,3)-glucan synthase (GS) which is a multimeric enzyme composed of a catalytic component and a regulatory component (the GTPase Rho1p). The aim of this paper is the characterization of new components that act above the path Rho1p/Pck1p/Pck2p that regulates cell integrity in Schizosaccharomyces pombe.
Mtl2p proteins and S. Wsc1p pombe structural characteristics similar to the "sensors" of S. Wsc1p and Mid2p cerevisiae. A single transmembrane domain, linking proteins with long extracellular region, a cytoplasmic domain at the carboxyl terminal, and a periplasmic ectodomain rich in serine and threonine. They are located on the cell surface but in different ways, Mtl2p is distributed throughout the cell periphery and the septum. Wsc1p is located so polarized and appears more focused on "small granules" at the poles of the cell and involved in regulating cell integrity where they shared a vital role. The simultaneous deletion and Wsc1 MTL2 + + is lethal. Wsc1p Mtl2p and participate in the activation of Rho1p and the two interact genetically Rgf1p (a GEF for Rho1p). Wsc1p is involved in regulating the biosynthesis of b (1,3)-glucan and probably also in a (1,3)-glucan. Wsc1p overexpressing cells show an overall increase in the total amount of cell wall deposition aberrant calcofluor. In addition, Wsc1p is necessary for activation of Rgf1p at least under stress conditions.
