<mods:abstract>[ES]Las GTPasas Ras controlan rutas de señalización implicadas en proliferación, migración, muerte&#xd;
y supervivencia celular, Actúan como interruptores moleculares, alternando entre una&#xd;
conformación inactiva (unido a GDP) y activa (unido a GTP), estando este proceso altamente&#xd;
regulado por proteínas activadoras de la actividad GTPasa intrínseca de Ras (GAPs, reguladores&#xd;
negativos), y factores de intercambio de nucleótidos de guanosina (GEFs, reguladores positivos).&#xd;
De entre las más de 150 GTPasas conocidas, la subfamilia de GTPasas Ras canonicas está&#xd;
constituida por HRas, NRas, y por las dos variantes Kras4A y Kras4B. A pesar de la gran&#xd;
homología, no son funcionalmente redundantes. Solo la pérdida individual de Kras4B, o la&#xd;
eliminación combinada de HRas y NRas junto con una haploinsuficiencia de Kras produce&#xd;
letalidad embrionaria. Sin embargo, hemos observado que la eliminación conjunta de HRas y&#xd;
NRas provocaba un aumento significativo de la letalidad perinatal, asociada con insuficiencia&#xd;
respiratoria. En esta Tesis Doctoral se ha llevado a cabo un análisis detallado de los modelos&#xd;
murinos knockouts (KO) para HRas y/o NRas con el fin de evaluar la especificidad o redundancia&#xd;
funcional de las dos GTPasas canónicas durante el desarrollo embrionario del pulmón</mods:abstract>
<mods:abstract>[EN]Ras GTPases are pivotal signal transduction molecules regulating cellular differentiation,&#xd;
proliferation, migration, apoptosis, and survival. They act as molecular switches by cycling&#xd;
between an inactive (GDP-bound) and active (GTP-bound) conformation in a process tightly&#xd;
modulated by GTPase activating proteins (GAPs, negative regulators) and Guanosine Exchange&#xd;
Factors (GEFs, positive regulators). Among the more than 150 known small GTPases, the&#xd;
canonical Ras subfamily includes HRas, NRas, as well as the two splicing isoforms, KRas4A and&#xd;
KRas4B. Despite their high similarity, the different Ras isoforms are not functionally redundant.&#xd;
Only the individual lack of KRas4B or the combined removal of HRas and NRas together with&#xd;
KRas haploinsufficiency result in embryonic lethality. Interestingly, we observed that&#xd;
HRas/NRas-devoid animalsshowed a significantly higher-than-expected mortality rate during the&#xd;
first hours after birth that was associated to severe respiratory distress. This dissertation presents&#xd;
an extensive analysis of knockout (KO) mouse models for HRas and/or NRas with an aim at&#xd;
uncovering specific or redundant functionalities of these two canonical Ras GTPases in lung&#xd;
embryonic development.</mods:abstract>
