<dc:description>[ES] Desde hace décadas, la búsqueda y el desarrollo de tratamientos eficaces para hacer&#xd;
frente a la pérdida neuronal de las enfermedades neurodegenerativas han supuesto un gran&#xd;
desafío para la ciencia y la sociedad. La mayoría de las terapias actuales son paliativas y&#xd;
están basadas en la filosofía “un gen, una enfermedad, un fármaco”. Sin embargo, existen&#xd;
pocos estudios en los que se haya explorado el efecto sinérgico resultante de la&#xd;
combinación de diferentes estrategias terapéuticas para impedir esta degeneración neuronal.&#xd;
Esta Tesis Doctoral analiza y compara las propiedades neuroprotectoras a nivel celular&#xd;
y comportamental de tratamientos combinados de una terapia farmacológica con Noleiletanolamina&#xd;
(OEA) y de terapia celular con células madre, en un modelo de&#xd;
degeneración neuronal cerebelosa: el ratón mutante PCD. En primer lugar, el tratamiento&#xd;
exclusivo con OEA retrasa las alteraciones morfológicas y la muerte de las neuronas en&#xd;
degeneración, modula la neuroinflamación y restaura funciones motoras, cognitivas y&#xd;
sociales alteradas en este modelo animal. En segundo lugar, los tratamientos combinados&#xd;
evaluados no muestran de forma global una neuroprotección sinérgica respecto al&#xd;
tratamiento farmacológico exclusivo con OEA, salvo en el comportamiento motor.&#xd;
Estos resultados ponen en evidencia la complejidad de la patología neurodegenerativa&#xd;
y de sus posibles tratamientos. Lejos de apoyar el enfoque empírico como solución a largo&#xd;
plazo, estos resultados invitan a una investigación más consolidada de los mecanismos&#xd;
subyacentes a estos fenómenos biológicos.&#xd;
[EN] For decades, the search and development of effective treatments to stop the defining&#xd;
neuronal loss of neurodegenerative diseases have posed a great challenge for science and society.&#xd;
Most of the current therapies are palliative and based on the “one gene, one disease, one drug” philosophy. However, few studies addressed the synergistic effect driven by the combination of&#xd;
different therapeutic strategies to prevent this neuronal degeneration.&#xd;
In this Doctoral Thesis, the neuroprotective properties of combined treatments of a&#xd;
pharmacological therapy with oleoylethanolamide (OEA) and stem cell therapy are analyzed and&#xd;
compared at the cellular and behavioral levels in a model of cerebellar neuronal degeneration: the&#xd;
PCD mutant mouse. First, exclusive treatment with OEA delays morphological alterations and&#xd;
death of degenerating neurons, modulates neuroinflammation, and restores impaired motor,&#xd;
cognitive, and social functions in this animal model. Secondly, the combined treatments assessed&#xd;
do not fully exert a synergistic neuroprotection with respect to the exclusive pharmacological&#xd;
treatment of OEA, except for motor behavior.&#xd;
These findings highlight the complexity of neurodegenerative pathology and its potential&#xd;
treatments. Far from supporting the empirical approach as a long-term solution, these results call&#xd;
for more consolidated research of the mechanisms underlying these biological phenomena.</dc:description>
