<dc:description>[ES] En esta Tesis Doctoral se pueden distinguir dos apartados diferentes y complementarios: &#xd;
síntesis de éteres lipídicos antitumorales análogos de miltefosina y perifosina y &#xd;
utilización del ácido ent-halímico como material de partida para la síntesis de &#xd;
compuestos bioactivos (neurotróficos, antiparasitarios y antitumorales).&#xd;
Dentro de la línea de investigación de síntesis de éteres lipídicos antitumorales, se han &#xd;
sintetizado análogos de miltefosina con insaturación en la cadena lipídica, yodados en &#xd;
mitad de cadena y funcionalizados al final de la cadena lipídica con función acetato, &#xd;
hidroxilo, éter y con un fluoróforo tipo BODIPY. También, se ha sintetizado un análogo &#xd;
de perifosina insaturada.&#xd;
Los análogos sintetizados se han ensayado como antitumorales frente a las líneas &#xd;
celulares HeLa y HL-60. Además, se han evaluado como antileishmaniosos frente a &#xd;
Leishmania major. El compuesto fluorescente 49 presenta excelentes propiedades &#xd;
espectroscópicas de absorción y emisión, haciéndolo interesante para estudios in vitro&#xd;
de la actividad antitumoral de los ATLs, a pesar de no presentar actividad antitumoral. &#xd;
El análogo de perifosina 25 presenta gran actividad como antitumoral en los ensayos &#xd;
realizados con la línea HL-60, mientras que no presenta actividad en HeLa. Cuando 25 se &#xd;
ensayó como antileishmanioso en L. major, se observó que es capaz de secuestrar el &#xd;
ciclo celular de L. major en fase sub G0/G1, siendo más activo que miltefosina.&#xd;
En la línea de investigación correspondiente a la síntesis de compuestos bioactivos a &#xd;
partir de ácido ent-halímico, se ha realizado la extracción del ácido ent-halímico, &#xd;
producto mayoritario del extracto en acetato de etilo de Halimium viscosum, como &#xd;
material de partida. Se han preparado una serie de compuestos de fácil acceso para &#xd;
ensayar su bioactividad como neurotróficos y como antiparasitarios frente a S. &#xd;
venezuelensis; sin observarse actividad para ninguna de las moléculas ensayadas.&#xd;
También, se ha avanzado en la aproximación sintética a ácido tessmánnico a partir de &#xd;
ácido ent-halímico. En esta síntesis se ha optado por una estrategia sintética que tiene &#xd;
las siguientes etapas fundamentales: funcionalización en C-2 y C-3, hidrogenación del &#xd;
doble enlace Δ1,10, apertura del anillo A y funcionalización de la cadena lateral. En &#xd;
trabajos futuros se terminará la síntesis del ácido tessmánnico, realizando cambios en &#xd;
la ruta sintética; primero rompiendo el anillo A y posteriormente realizando la &#xd;
hidrogenación.&#xd;
[EN] This Thesis can be divided in two different and complementary research lines: Synthesis &#xd;
of antitumoral ether lipids analogues of miltefosine and perifosine and the use of ent halimic acid as starting material for the synthesis of bioactive compounds (neurotrophic, &#xd;
antiparasitary and antitumoral).&#xd;
Within the research line of antitumoral ether lipids, analogues of miltefosine with &#xd;
unsaturation and iodine in the middle position of the lipid chain as well as functionalized &#xd;
at the end of the lipid chain (with acetate, hydroxyl, ether and BODIPY-type fluorophore) &#xd;
were synthesized. An analogue of perifosine with unsaturation in the middle position of &#xd;
the carbon chain was also prepared.&#xd;
The synthesized analogues have been assayed as antitumor agents vs HeLa and HL-60 &#xd;
cell lines. In addition, compound 25 was evaluated as antileishmanial testing the activity &#xd;
in Leishmania major. The fluorescent compound 49 has excellent absorption and &#xd;
emission spectroscopic properties, opening the possibility of its application for in vitro &#xd;
studies of the antitumor activity of ATLs, despite not showing remarkable antitumoral &#xd;
activity so far.&#xd;
The perifosine analogue 25, shows great antitumoral activity against HL-60 cell line, &#xd;
while does not show activity against HeLa. The antileishmanial assays of 25 against L. &#xd;
major presented the unsaturated derivative 25 as a more potent antileishmanial &#xd;
compared with miltefosine, arresting the cell cycle of the parasite in sub G0/G1 phase.&#xd;
In the research line corresponding to the synthesis of bioactive compounds using ent halimic acid as starting material, it was carried out the extraction with ethyl acetate of &#xd;
Halimium viscosum isolating its major compound: ent-halimic acid. A series of facile &#xd;
accessible compounds where synthesized and evaluated as antiparasitary against S. &#xd;
venezuelensis and neurotrophic, but no activity was observed so far.&#xd;
In this work, it was also afforded a synthetic approach to tessmannic acid starting from &#xd;
ent-halimic acid. In this synthetic route, it was tried a synthetic strategy consisting on: &#xd;
the functionalization at C-2 and C-3, hydrogenation of the Δ1,10 double bond, ring-A &#xd;
cleavage, and functionalization of the side chain. In a continuation of this work, a &#xd;
different synthetic strategy will be tried; affording firstly the ring-A opening and then, &#xd;
the hydrogenation of the double bond.</dc:description>
