<mods:abstract>[EN] Centrosomes are the major microtubule-organizing centres in eukaryotic cells. In mitosis and male meiosis, centrosomes&#xd;
nucleate and organize the microtubules that build up the spindle, the macromollecular apparatus that generates and&#xd;
guides the physical forces needed to segregate chromosomes into daughter cells. Contrarily, in female meiosis of most&#xd;
metazoa, the female progenitor cell actively eliminate centrosomes before or during meiotic divisions. Consequently, the&#xd;
meiotic spindle assembles and segregates chromosomes without the guidance of centrosomes (‘acentrosomal spindle’).&#xd;
The fission yeast Schizosaccharomyces pombe is excellent to study meiosis, although it maintains the SPBs (fungal equivalent&#xd;
of centrosomes) in meiosis and so, the meiotic spindle is still mediated by the SPBs in this organism (‘SPB-mediated&#xd;
spindle’). This difference with female metazoan meiosis limits the potential of this yeast for studying the molecular basis&#xd;
underlying the acentrosomal spindle, interesting to understand the meiotic errors affecting human fertility and health.&#xd;
In our lab, we generated a fission yeast system (bqt1Δ sad1.2) where SPBs are dislodged from the nuclear envelope, so&#xd;
that the SPB-dependent spindle formation is abolished. Here, we observe a microtubule array with spindle-like morphology&#xd;
and dynamics and chromosome segregation capability despite the dislodgement of SPBs, hypothesizing that&#xd;
this structure is a self-assembled spindle, independent of SPBs in fission yeast meiosis in analogy with the acentrosomal&#xd;
spindle independent of centrosomes in female metazoan meiosis. In this doctoral thesis, we explore the possibility of&#xd;
such type of spindle, optimizing the experimental conditions for its formation and study. We confirm the formation of&#xd;
the self-assembled spindle independently of the SPBs and validate this by reproducing it through alterative genotypes.&#xd;
Also, we describe that its assembly depends on the microtubule polymerizer and nucleator Alp14/XMAP215 and that&#xd;
the microtubule crosslinker Ase1/PRC1 is necessary to maintain its structural integrity during elongation. However,&#xd;
its formation is majorly independent of the γ-tubulin complex activity, instead depending on mechanisms conserved in&#xd;
metazoa, crutial for acentrosomal spindle formation, such as the kinetochore pathway and the RanGTP gradient. Further,&#xd;
we improve its structural robustness and fidelity of chromosome segregation by stabilizing microtubules via the&#xd;
elimination of Kinesin-8, especifically Klp6 but not Klp5. Lastly, we standarized our methodology for self-assembled&#xd;
spindle obention, observation through fluorescence microscopy timelapse and analysis, enabling a deeper characterization&#xd;
of spindle assembly, elongation dynamics and chromosome segregation.&#xd;
[ES] Los centrosomas son los principales centros organizadores de microtúbulos en las células eucariotas. En mitosis y meiosis&#xd;
masculina, los centrosomas nuclean y organizan los microtúbulos que construyen el huso, el aparato macromolecular&#xd;
que genera y guía las fuerzas físicas necesarias para segregar los cromosomas en las células hijas. Por el contrario, en la&#xd;
meiosis femenina de la mayoría de metazoos, la célula progenitora femenina activamente elimina los centrosomas antes&#xd;
o durante las divisiones meióticas. Consecuentemente, el huso meiótico se ensambla y segrega cromosomas en ausencia&#xd;
de la guía de los centrosomas (‘huso acentrosómico). La levadura de fisión Schizosaccharomyces pombe es excelente para&#xd;
estudiar la meiosis, aunque esta mantiene los SPBs (equivalentes fúngicos de los centrosomas) en meiosis y, entonces, el&#xd;
huso meiótico sigue siendo mediado por los SPBs en este organismo (’huso dependiente de SPB’). Esta diferencia con la&#xd;
meiosis femenina de metazoos limita el potencial de esta levadura para estudiar la base molecular subyacente al huso&#xd;
acentrosómico, interesante para entender los errores meioóticos que afectan a la fertilidad y salud humanas.&#xd;
En nuestro laboratorio, hemos generado un sistema de levadura de fisión (bqt1Δ sad1.2) en el que los SPBs están aislados&#xd;
de la envoltura nuclear, de tal forma que la formación huso dependiente de SPBs está abolida. Aquí, observamos un&#xd;
array de microtúbulos con una morfología y dinámicas típicas de un huso y capacidad de segregación de cromosomas&#xd;
a pesar del aislamiento de los SPBs, hipotetizando que esta estructura es un huso autoensamblado, independiente de&#xd;
los SPBs en la meiosis de la levadura de fisión, análogo al huso acentrosómico independiente de los centrosomas en la&#xd;
meiosis femenina de metazoos. En esta tesis doctoral, exploramos la posibilidad de dicho tipo de huso, optimizando&#xd;
las condiciones experimentales para su formación y estudio. Confirmamos la formación del huso autoensamblado independientemente&#xd;
de los SPBs y validamos esto reproduciéndola mediante genotipos alternativos. También, describimos&#xd;
que su ensamblaje depende del polimerizador y nucleador de microtúbulos Alp14/XMAP215 y que el entrecruzador de&#xd;
microtúbulos Ase1/PRC1 es necesario para mantener su integridad estructural durante su elongación. Sin embargo, su&#xd;
formación es mayormente independiente de la actividad del complejo de γ-tubulina y, en su lugar, depende de mecanismos&#xd;
conservados en metazoos, cruciales para la formación del huso acentrosómico, como la ruta del quinetocoro y&#xd;
el gradiente de RanGTP. Más allá, mejoramos su robustez estructural y fidelidad de segregación de cromosomas con&#xd;
la eliminación de la Quinesina-8, específicamente Klp6 pero no Klp5. Por último, estandarizamos nuestra metodología&#xd;
para la obtención, observación por videomicroscopía de fluorescencia y análisis del huso autoensamblado, lo cual permitió&#xd;
una caracterización más en detalle de su ensamblaje, dinámica de elongación y segregación de cromosomas.</mods:abstract>
