[EN]Paclitaxel is an anticancer drug used in the treatment of ovarian, breast, non small cell lung, and Kaposi’s sarcoma associated with AIDS. Its mechanism of action is based on blocking β-tubulin subunit of microtubules, decreasing depolymerisation and stabilizing them; stopping the cell cycle at the G2 / M phase leading to apoptosis. Furthermore, the autophagy, in which process microtubules are very important, is a set of catabolic processes crucial for the maintenance of cell viability; but also is a programmed cell death. The target of this article is to study the drug cytotoxicity and its effect on 6 autophagy related proteins: P62, PKC ζ, mTOR, TRAF6, LC3 and Beclin 1; on two colon cancer cell lines: HT29 and HCT-116. The techniques used were the MTT and Western Blot tests. Our results indicate that paclitaxel inhibits the growth of both lines at 10 nM; and increases the expression of Beclin 1, LC3-II and p62 proteins, which indicates an induction of autophagy, and a later block of autolisosoma. In conclusion, we can say that paclitaxel blocks autophagy. Key words: paclitaxel; cáncer; colon; autophagy; p62; LC3-II; Beclin 1.
[ES]El paclitaxel es un fármaco antitumoral utilizado en el tratamiento de cáncer de ovario, mama, no microcítico de pulmón y sarcoma de Kaposi asociado al SIDA. Su mecanismo de acción se basa en el bloqueo de la subunidad β-tubulina en el microtúbulo, disminuyendo su despolimerización y estabilizándolos; deteniendo el ciclo celular en la fase G2/M conduciendo a la apoptosis. Por otro lado, la autofagia, donde tienen gran importancia los microtúbulos, es un conjunto de procesos catabólicos cruciales para el mantenimiento de la viabilidad celular; pero también supone una ruta de muerte celular programada. Con este trabajo se pretende estudiar la citotoxicidad de este fármaco, así como su efecto sobre 6 proteínas relacionadas con la autofagia: P62, PKC ζ, mTOR, TRAF6, LC3 y Beclin 1; sobre dos líneas celulares de cáncer de colon: HT29 y HCT-116. Las técnicas utilizadas fueron los ensayos MTT y Western Blot. Nuestros resultados indican que el paclitaxel inhibe el crecimiento de ambas líneas a 10nM; y aumenta la expresión de las proteínas Beclin 1, LC3-II y p62, que nos indica una inducción de la autofagia, y su posterior bloqueo en el paso de formación del autolisosoma. En conclusión, se puede afirmar que el paclitaxel bloquea la autofagia.
