Implicación de promotores y transportadores de aniones orgánicos de las familias OATP y ABC en patología, toxicología y terapia antitumoral

[ES] En los hepatocitos, los procesos de captación desde la sangre portal de fármacos y
toxinas, así como de numerosos compuestos endógenos como ácidos biliares (ABs) y
bilirrubina están mediados por diversas familias de transportadores. Entre ellas, la más
importante por sus implicaciones y número de miembros es la familia de proteínas
OATP (Organic Anion Transporting Polypeptide) codificadas por los genes SLCO. Por
otra parte, la expulsión de sustancias orgánicas mediante transporte activo primario a
la bilis en situaciones fisiológicas, o de vuelta hacia la sangre cuando su salida hacia la
bilis está dificultada en situaciones de colestasis se debe en gran medida a miembros
de la superfamilia de proteínas ABC (ATP-Binding Cassette).
Un mejor conocimiento de estos procesos puede ser de gran utilidad para la
manipulación farmacológica de la vectorización de fármacos, para prevenir la
captación o mejorar la eliminación de tóxicos, o para comprender la etiopatogenia de
trastornos relacionados con la dinámica de sustancias endógenas que son sustratos
de estos transportadores.
Aprovechando la analogía de la metodología necesaria para su estudio, el
conocimiento de estos sistemas de transporte por parte de nuestro grupo de
investigación, y la actualidad de diversas cuestiones pendientes de respuesta en este
campo, nos planteamos como objetivo general de esta Tesis Doctoral profundizar
en el estudio del papel que juegan estas proteínas y los mecanismos que regulan su
expresión en diferentes aspectos de la Fisiología, la Fisiopatología y la Farmacología.
Este objetivo general engloba los siguientes tres objetivos parciales que nos fuimos
planteando a lo largo del periodo de formación predoctoral:
Objetivo 1º: Investigar la capacidad de varios derivados de ABs para inhibir la
captación mediada por proteínas de la familia OATP. El interés de este objetivo era
evaluar la utilidad de bloqueantes de transportadores OATP como antídotos en casos
de consumo accidental de toxinas captadas por estas proteínas. Un ejemplo es la
faloidina presente en setas venenosas, como la Amanita phalloides.
Objetivo 2º: Investigar el papel de transportadores ABC en la etiopatogenia del
síndrome respiratorio neonatal asociado a la hipercolanemia materna que acompaña a
la colestasis de la embarazada.
Objetivo 3º: Investigar los mecanismos que controlan la sobreexpresión de proteínas
ABC y que reducen la respuesta de las células de cáncer de colon a la quimioterapia.
Como fármaco modelo se eligió el cisplatino y los estudios se centraron en la proteína
ABCC2, que se cree que juega un papel importante en la resistencia a este citostático.

to study the role of these proteins and the mechanisms that regulate their
expression in different aspects of Physiology, Pathophysiology and Pharmacology.
This overall objective includes the following three sub-goals that we went
raising over predoctoral training period:
Objective 1: To investigate the ability of various derivatives of ABs to inhibit
mediated uptake by OATP family proteins. The interest of this objective was
evaluate the usefulness of OATP transporter blockers as antidotes in cases
accidental consumption of toxins captured by these proteins. An example is the
phalloidin present in poisonous mushrooms such as Amanita phalloides.
Objective 2: To investigate the role of ABC transporters in the pathogenesis of
neonatal respiratory syndrome associated with maternal hipercolanemia accompanying
cholestasis of pregnancy.
Objective 3: To investigate the mechanisms that control protein overexpression
ABC and reduce the response of colon cancer cells to chemotherapy.
Was chosen as a model drug cisplatin and the studies focused on protein
