Resumen
[ES] La presente tesis trata sobre las alteraciones en células secretoras del páncreas exocrino en modelos experimentales de fibrosis quística y pancreatitis aguda. En una primera parte se realiza una introducción a la secreción pancreática exocrina, pancreatitis aguda y fibrosis quística. Los objetivos de este trabajo son investigar si la respuesta secretora al incremento en la concentración de cAMP depende de la activación de la PKA en los conductos pancreáticos de ratones normales y CF; investigar si la activación de la ruta intracelular iniciada por la unión del cAMP a las proteínas Epac está implicada en la regulación de los fenómenos secretores en conductos pancreáticos del ratón; e investigar el mecanismo que mantiene la secreción ductular en ratones CF, mediante el estudio de los cambios en la conductancia aniónica inducidos por el cAMP en células ductulares pancreáticas. Finalmente se desarrollan una hipótesis y objetivos, además de una metodología, y por último se ofrecen los resultados obtenidos y las cuestiones que quedan abiertas a discusión. [EN] This thesis discusses the changes in secretory cells of the exocrine pancreas in experimental models of severe pancreatitis and cystic fibrosis. The aims of this study are to investigate if the secretory response to increased cAMP concentration depends on PKA activation in the pancreatic ducts of normal mice and CF; investigate if the intracellular route activation initiated by the union of cAMP and Epac proteins is involved in regulating the secretory phenomena in mouse pancreatic ducts; and investigate the mechanism that maintains the ductular secretion in CF mice, through the study of changes in anionic conductance induced by cAMP in pancreatic ductulares cells. Finally, develop a hypothesis and objectives, as well as a methodology, and finally gives the results obtained and the questions that remain open to discussion.
